The molecular mechanism by which thymocytes are positively selected remains incompletely understood. Three studies add a new piece to the positive selection puzzle. Tcell antigen receptor (TCR) signaling has been intensively studied over the past 25 years. After recognition of peptide-major histocompatibility complex by the TCR, a proximal ordered cascade of signaling events occurs involving CD3 chains, the tyrosine kinases Lck and Zap70, and adaptor molecules, including Lat and Grb2. The present textbook models of TCR signaling are based on the assumption that most, if not all, of the molecules critical for TCR signaling have been identified. But surprises still occur in science: in this issue, three separate groups identify a previously unknown molecule, called 'Themis' (for 'thymus-expressed molecule involved in selection'), that is critical in the positive selection of developing T cells.
