摘要:
目的 探究基质金属蛋白酶-2(matrix metalloproteinase -2,MMP-2)、凋亡抑制基因(Survivin)、白介素-6(interleukin-6,IL-6)、信号传导及转录活化因子3(signal transducers and activators of transcription 3, STAT 3)在子痫前期患者胎盘中的水平及意义.方法 选取乐陵市人民医院2016年3月至2017年5月收治的76例子痫前期孕妇为研究对象,按照病情分为轻度子痫前期组(34例)与重度子痫前期组(42例),另选取同期35例正常孕妇为对照组,分娩后收集胎盘组织,HE染色观察各组胎盘组织变化,免疫组化法检测胎盘组织中MMP-2、Survivin、IL-6和STAT 3表达,Pearson相关性分析MMP-2、Survivin、IL-6、和STAT 3与临床指标的相关性.结果 子痫前期组收缩压、舒张压、尿蛋白均高于对照组,新生儿体重低于对照组,且随着病情程度加重,重度子痫前期组变化程度更为显著(P<0.05);免疫组化显示,MMP-2、Survivin、STAT 3在3组中阳性表达量、平均光密度逐渐降低,IL-6阳性表达量、平均光密度逐渐增加( P<0.05);收缩压、舒张压、尿蛋白与MMP-2、Survivin、STAT 3呈负相关,与IL-6呈正相关;新生儿体重与MMP-2、Survivin、STAT 3呈正相关,与IL-6呈负相关(P<0.05).结论 MMP-2、Survivin、STAT 3低表达,IL-6高表达与子痫前期发生发展、新生儿的预后有关.%Objective To explore the levels and significance of matrix metalloproteinase -2 ( MMP-2),apoptosis inhibitor gene(Survivin),interleukin-6(IL-6),signal transducer and activator of transcription 3(STAT 3) in placenta of patients with preeclampsia.Methods 76 cases of pre-eclampsia pregnant women from March 2016 to May 2017 in Leling People's Hospital were selected as the study subjects.According to the condition, 34 cases were in the mild preeclampsia group ,42 cases in the severe preeclampsia group,and 35 normal pregnant woman were the control group.Placental tissues were collected after delivery , and the changes of placental tissue were observed by HE staining.The expressions of MMP-2, Survivin, IL-6 and STAT 3 in placenta were detected by immunohistochemistry.Pearson correlation analysis was performed on MMP-2, Survivin and IL-6, STAT 3 and the correlation with clinical indicators.Results The systolic blood pressure, diastolic blood pressure and urine protein in the preeclampsia group were higher than those in the control group.The neonatal weight was lower than that in the control group , and the severity of the severe preeclampsia group was more significant ( P<0.05).Immunohistochemistry showed that the positive expression and average optical density of MMP-2, Survivin and STAT 3 decreased gradually, and the positive expression of IL-6 and the average optical density increased gradually (P<0.05).Systolic blood pressure, diastolic blood pressure, urine protein were significantly negatively correlated with MMP-2, Survivin and STAT 3, and positively correlated with IL-6.Neonatal weight was significantly positively correlated with MMP-2, Survivin and STAT 3, and IL-6 showed a significant negative correlation (P<0.05).Conclusion Low expression of MMP-2, Survivin and STAT 3, high expression of IL-6 is associated with the development of preeclampsia and the prognosis of neonates .
