    • 摘要: 大曲糖化力是评价大曲质量的一项重要指标,但常规所采用的斐林试剂滴定法影响因素较多.采用3,5-二硝基水杨酸作为指示剂,并对测定波长、加热条件进行优化,建立了一种糖化力的快速检测方法,本方法快速准确科学,可用于大曲糖化力的检测及应用.

    • 摘要: 目的 探究自酿原浆啤酒发酵过程中蛋白质和总糖含量的变化.方法 采用山西大同大学生物工程试验中心酿造的啤酒为原料,采用考马斯亮蓝比色法和3,5二硝基水杨酸(DNS)定糖法测定其蛋白质和总糖含量,并与市售常见燕京啤酒、哈尔滨啤酒和纯生啤酒进行分析比较.结果 麦芽醪在整个发酵过程中,蛋白质含量处于变化中.第9 d和第13 d的蛋白含量为发酵期间的两个极大值,约为2.0 mg/mL;临近出酒时,蛋白质含量在1.5 mg/mL上下波动.总糖量前4d急剧下降,从94.2 mg/mL下降到42.8 mg/mL,后10d总糖含量在6.7 mg/mL上下波动;出酒时的含量约为2.7 mg/mL.自酿原浆啤酒蛋白质和总糖含量均高于市售常见啤酒的含量.结论 原浆啤酒发酵过程中蛋白质和糖的变化具有规律性,可以作为改良啤酒酿造工艺的参考依据.

    • 摘要: 建立了大米中水溶性糖的测定方法.大米中的水溶性糖用热水提取,乙酸锌、亚铁氰化钾溶液沉淀,经盐酸水解后用3,5-二硝基水杨酸显色,分光光度计比色测定.结果 表明,在3,5二硝基水杨试剂用量1.0 mL,沸水浴加热6 min,540 nm波长条件下进行测定,标准曲线线性关系良好,回归方程Y=0.968 2X+0.014 4,相关系数R2=0.999 6,在0.1%、0.5%和2.0%三个标准添加浓度下,水溶性糖的平均加标回收率为91.9% ~98.3%,相对标准偏差为2.68% ~ 2.79%,方法检出限为0.003%.该方法准确度和灵敏度高,重现性好,操作简单、快速,有应用价值.10份不同品种大米水溶性糖含量测定结果为0.12% ~1.95%.

    • 摘要: 方法基于在碱性条件下,葡萄糖与3,5-二硝基水杨酸作用产生的棕红色物质在540nm处有较大的吸光度,在一定浓度范围内,葡萄糖的量与棕红色物质颜色的深浅成比例关系.通过实验,确定了最佳条件:最佳测量波长为540nm,显色剂用量为1.5g,显色时长最佳为2min.该方法操作简单易行,结果准确率高,重复性好.

    • 摘要: Two new complexes based on 3,5-dinitrosalicylic acid (H2dns) and 1,10-phenanthroline (phen),namely [La2(dns)2(Hdns)2(phen)4] (1) and [La4(dns)6(phen)d (2) have been synthesized and characterized by IR,elemental analyses,thermogravimetric analyses and X-ray diffraction technique.Single-crystal X-ray diffraction analyses revealed that two complexes all crystallize in the triclinic system and space group P(1).For complexes 1,H2dlns ligands adopt tridentate and didentate coordination modes to link La(Ⅲ) ions into dinuclear structure.However H2dns ligands adopt tridentate coordination mode to link La (Ⅲ) ions into tetranuclear structure.TG and fluorescence analysis show the thermal stability of 2 is higher than that of 1,and 1 and 2 have same maximum emission band of the fluorescence at 467 nm.CCDC:1856530,1;1856531,2.%以3,5-二硝基水杨酸(H2dns)和菲咯啉(phen)为配体合成了2个新的镧配合物[La2(dns)2(Hdns)2(phen)4] (1)和[La4(dns)6(phen)6](2),并对它们进行了红外分析、元素分析、热重分析和单晶结构分析.单晶衍射结果表明,配合物1和2均为三斜晶系,Pi空间群.配合物1和2中La(Ⅲ)通过配体H2dns桥联分别形成双核结构和四核结构.热重和荧光分析表明:2热稳定性高于1,1和2均在467 nm处有最强荧光发射强度.

    • 摘要: 研究重离子7Li辐射对“永19”玉米自交系还原糖含量的影响.采用溴化钾压片法红外光谱定性表征玉米还原糖.优化3, 5-二硝基水杨酸(DNS)法的最佳波长,在540 nm定量测定不同剂量重离子7Li辐射自交系的还原糖含量.提取液与标准品特征峰重合证明提取液为还原糖.定量结果表明10 Gy辐射还原糖含量高达4.50%,20和40 Gy辐射分别为3.94%、3.79%,而30和50 Gy辐射略低于对照(3.55%).重离子7Li辐射对玉米还原糖含量有正负双重作用呈现出马鞍形曲线.%An investigation demonstrated the effects of heavy ion 7Li irradiation on "Yong 19" maize inbred lines' reducing sugar content.Using KBr tableting technique,maize reducing sugar was qualitatively characterized by infrared spectrum.Then,optimized the wavelength for 3,5-dinitro salicylic acid (DNS) determination,all the reducing sugars from different doses of heavy ion 7Li treated inbred lines were quantitatively measured at 540 nm.These characteristic peaks of extraction and standard were coincident,which proved that the extraction was reducing sugar.The quantitative results showed that reducing sugar content of 10 Gy radiation reached as high as 4.50%,20 and 40 Gy radiation were 3.94% and 3.79%,whereas 30 and 50 Gy radiation were slightly lower than that of the control sample (3.55%).It indicated that heavy ion 7Li irradiation has both positive and negative effects on maize reducing sugar content,which presented a kind of saddle-type curve.

    • 摘要: 3 ,6-Dihydrazine-1 ,2 ,4 ,5-tetrazine- 3 ,5-dinitrosalicylic acid salt dihydrate (DHT · 2DNS · 2H2O ) was synthesized using 3 ,6-dihydrazine-1 ,2 ,4 ,5-tetrazine (DHT ) and 3 ,5-dinitrosalicylic acid (DNS ) as raw materials and its structure was characterized by nuclear magnetic resonance spectroscopy ,infrared spectroscopy ,element a-nalysis and thermogravimetry . The thermal behavior and thermal decomposition products of anhydrous DHT · 2DNS were studied by differential scanning calorimetry (DSC) and thermogravimetry combined with Fourier trans-form infrared analysis (TG-FTIR) . The constant-volume combustion heat (Δc U ) was determined by an oxygen bomb calorimeter and the standard molar enthalpy of combustion (Δc H ㈠m ) and the standard molar enthalpy of forma-tion (Δf H ㈠m ) were estimated via the Δc U .The results show that the thermal stability of DHT · 2DNS is higher than DHT and its thermal decomposition process can be divided into four stages .Firstly ,part of hydroxyl ,nitro and car-boxyl in DNS ion of DHT · 2DNS were removed and hydrazine was decomposed ;then ,the tetrazine ring and groups on the benzene ring was dissociated ,thirdly ,benzene ring was broken ;finally ,the benzene residues kept decompo-sing .The values of Δc U ,Δc H ㈠m and Δf H ㈠m of DHT · 2DNS are ( - 11938 .17 ± 25 .33)J/g ,( - 7119 .77 ± 15 .16)kJ/mol and ( - 1177 .2 ± 11 .14)kJ/mol ,respectively .%以3,6-二肼基四嗪(DHT)和3,5-二硝基水杨酸(DNS)为原料,合成了3,6-二肼基-1,2,4,5-四嗪二硝基水杨酸盐二水合物(DHT·2DNS·2H2 O),采用核磁共振波谱 、红外光谱 、元素分析及热重法对其结构进行了表征.采用差示扫描量热法(DSC)并结合热重-红外联用(TG-FTIR)对无水DHT·2DNS的热行为及热分解产物进行了研究.利用氧弹量热仪测定了DHT·2DNS的恒容燃烧热(Δc U),并通过 Δc U估算了标准摩尔燃烧焓(Δc H㈠m)和标准摩尔生成焓(Δf H㈠m).结果表明,DHT·2DNS的热稳定性要高于DHT,其受热分解可分为4个阶段:首先DHT·2DNS的二硝基水杨酸离子脱去羟基和部分硝基 、羧基,同时肼基发生分解;然后苯环上的基团与四嗪环发生解离;随后苯环发生裂解;最后苯环残余物继续发生分解.DHT·2DNS的 Δc U值 、Δc H㈠m值和 Δf H㈠m值分别为(-11938.17±25.33)J/g、(-7119.77±15.16)kJ/mol和(-1177.2±11.14)kJ/mol.

    • 摘要: 衣物上常见的淀粉类污垢有巧克力、土豆泥、面条、粥等,加入淀粉酶的洗衣液对去除这类污垢效果良好;另外织物表面因多次洗涤而在主纤维上出现微毛和小绒球,纤维素酶可将主纤维上的微毛和绒球除去,从而使主纤维变得光滑柔软,并具有增白效果.本实验选取革兰氏阳性菌和革兰氏阴性菌的两种代表菌(藤黄八叠球菌和大肠杆菌),通过牛津杯法测定消毒型洗衣液对上述两种菌的抑菌效果.结果显示,该消毒型洗衣液对藤黄八叠球菌(革兰氏阳性菌)有抑菌效果,而对大肠杆菌(革兰氏阴性菌)无抑菌作用.同时,以中温淀粉酶和中温纤维素酶样品作为研究对象,通过DNS法测定加入洗衣液前后相应酶的活力,结果显示加入1%洗衣液后淀粉酶的活力降低32.8%,纤维素酶的活力降低5.4%,这说明消毒型洗衣液中的成分对上述两种酶的活力有一定的影响,从而影响酶在洗涤方面的应用.%For the common starchy dirt (chocolate,mashed potatoes,noodles and porridge) on clothing,laundry detergent which contains amylase can remove dirt effectively.While micro-hair and small pompon appear on the main fibers due to repeated washing of the fabric surface,the cellulase in laundry detergent can remove micro-hair and small pompon on the primary fibers,then,the main fibers become smooth and soft,while it has whitening effect.In this study,two representatives of Gram-positive bacteria and Gram-negative bacteria (Sarcina lutea and Escherichia coli) were selected,antibacterial effect for the two-type bacteria of disinfection type of laundry detergent was determined by using Oxford cup method,the results showed that the disinfectant type of laundry detergent had antibacterial effects on Sarcina lutea (Gram-positive bacteria),however no inhibitory effect on Escherichia coli(Gram-negative bacteria).Besides,the mesophilic amylase and cellulase samples were applied for the research,the impact of enzyme activities of two enzymes generated by adding laundry detergent was determined by DNS colorimetry assay.It turned out that enzyme activities of the mesophilic amylase and cellulase reduced 32.8% and 5.4% after adding one percent of laundry detergent,respectively.The conclusions of the study showed that compositions of disinfectant laundry detergent had certain effects on the enzyme activities of amylase and cellulase in detergent industry.

    • 摘要: 目的:比较不同产地浙贝母的酸性磷酸脂酶(ACP)和淀粉酶(AMY)活性.方法:采用对硝基酚比色法测定ACP活性,采用3,5-二硝基水杨酸比色法测定AMY活性,采用聚类分析方比较研究不同产地浙贝母ACP和AMY活性.结果:不同产地浙贝母的ACP和AMY活性存在较大差异,磐安冷水产的浙贝母两酶活性最高,而江苏浙贝母两酶最低.结论:ACP和AMY活性比较研究可作为浙贝母来源鉴定的依据之一.

    • 摘要: 目的:建立了使用3,5-二硝基水杨酸比色法测定甘草多糖中多糖的含量.方法:将配置好的3,5-二硝基水杨酸显色液与经简单处理后达到设定浓度的对照品溶液和供试品溶液反应,取反应后生成的黄色溶液置于紫外分光光度仪中在520nm的波长处测定,分别测定单糖与总糖的含量,进而求出多糖的含量.结果:线性范围100 ~ 420μgr=0.9999,平均回收率100.8%,RSD=1.4%.结论:本方法简单、准确、重现性好,适用于含有多糖的药品或保健品的质量检查验.
